    Nuclear Fuel Handling Device

    The fuel handling device project is a solution to help the operators at Rhode Island Nuclear Science Center and Los Alamos National Laboratories ease the transportation and inspection of spent fuel rods. The objective is to have a device or pole that can remove spent fuel assemblies which sit in a 35 foot deep reactor. Which latches onto a small crossbar on top of the fuel assembly which transports it to another side of the reactor. In order to make sure it’s transported safely, there can’t be any errors where the fuel assembly would fall over. A device was created to meet the criteria that was laid out by the workers at RINSC and provide an optimal device for the workers to use. After 120 conceptual ideas and many hours of discussing these options, a final design has been chosen for the device. This includes specific dimensions and materials acceptable for use while also deciding to have an interchangeable end for multi use. Parts have been designed and printed to visualize the optimal end for the fuel handling before purchasing and manufacturing the final product

    Pivot Tracing: Dynamic Causal Monitoring for Distributed Systems

    Abstract Monitoring and troubleshooting distributed systems is notoriously di cult; potential problems are complex, varied, and unpredictable. e monitoring and diagnosis tools commonly used today -logs, counters, and metrics -have two important limitations: what gets recorded is de ned a priori, and the information is recorded in a component-or machine-centric way, making it extremely hard to correlate events that cross these boundaries. is paper presents Pivot Tracing, a monitoring framework for distributed systems that addresses both limitations by combining dynamic instrumentation with a novel relational operator: the happened-before join. Pivot Tracing gives users, at runtime, the ability to de ne arbitrary metrics at one point of the system, while being able to select, lter, and group by events meaningful at other parts of the system, even when crossing component or machine boundaries. We have implemented a prototype of Pivot Tracing for Java-based systems and evaluate it on a heterogeneous Hadoop cluster comprising HDFS, HBase, MapReduce, and YARN. We show that Pivot Tracing can e ectively identify a diverse range of root causes such as so ware bugs, miscon guration, and limping hardware. We show that Pivot Tracing is dynamic, extensible, and enables cross-tier analysis between inter-operating applications, with low execution overhead

    Retro: Targeted Resource Management in Multi-tenant Distributed Systems

    Abstract In distributed systems shared by multiple tenants, effective resource management is an important pre-requisite to providing quality of service guarantees. Many systems deployed today lack performance isolation and experience contention, slowdown, and even outages caused by aggressive workloads or by improperly throttled maintenance tasks such as data replication. In this work we present Retro, a resource management framework for shared distributed systems. Retro monitors per-tenant resource usage both within and across distributed systems, and exposes this information to centralized resource management policies through a high-level API. A policy can shape the resources consumed by a tenant using Retro's control points, which enforce sharing and ratelimiting decisions. We demonstrate Retro through three policies providing bottleneck resource fairness, dominant resource fairness, and latency guarantees to high-priority tenants, and evaluate the system across five distributed systems: HBase, Yarn, MapReduce, HDFS, and Zookeeper. Our evaluation shows that Retro has low overhead, and achieves the policies' goals, accurately detecting contended resources, throttling tenants responsible for slowdown and overload, and fairly distributing the remaining cluster capacity

    Isolation and characterization of novel microsatellite markers and their application for diversity assessment in cultivated groundnut (Arachis hypogaea)

    <p>Abstract</p> <p>Background</p> <p>Cultivated peanut or groundnut (<it>Arachis hypogaea </it>L.) is the fourth most important oilseed crop in the world, grown mainly in tropical, subtropical and warm temperate climates. Due to its origin through a single and recent polyploidization event, followed by successive selection during breeding efforts, cultivated groundnut has a limited genetic background. In such species, microsatellite or simple sequence repeat (SSR) markers are very informative and useful for breeding applications. The low level of polymorphism in cultivated germplasm, however, warrants a need of larger number of polymorphic microsatellite markers for cultivated groundnut.</p> <p>Results</p> <p>A microsatellite-enriched library was constructed from the genotype TMV2. Sequencing of 720 putative SSR-positive clones from a total of 3,072 provided 490 SSRs. 71.2% of these SSRs were perfect type, 13.1% were imperfect and 15.7% were compound. Among these SSRs, the GT/CA repeat motifs were the most common (37.6%) followed by GA/CT repeat motifs (25.9%). The primer pairs could be designed for a total of 170 SSRs and were optimized initially on two genotypes. 104 (61.2%) primer pairs yielded scorable amplicon and 46 (44.2%) primers showed polymorphism among 32 cultivated groundnut genotypes. The polymorphic SSR markers detected 2 to 5 alleles with an average of 2.44 per locus. The polymorphic information content (PIC) value for these markers varied from 0.12 to 0.75 with an average of 0.46. Based on 112 alleles obtained by 46 markers, a phenogram was constructed to understand the relationships among the 32 genotypes. Majority of the genotypes representing subspecies <it>hypogaea </it>were grouped together in one cluster, while the genotypes belonging to subspecies <it>fastigiata </it>were grouped mainly under two clusters.</p> <p>Conclusion</p> <p>Newly developed set of 104 markers extends the repertoire of SSR markers for cultivated groundnut. These markers showed a good level of PIC value in cultivated germplasm and therefore would be very useful for germplasm analysis, linkage mapping, diversity studies and phylogenetic relationships in cultivated groundnut as well as related <it>Arachis </it>species.</p

    Sipha maydis sensitivity to defences of Lolium multiflorum and its endophytic fungus Epichloë occultans

    Background. Plants possess a sophisticated immune system to defend from herbivores. These defence responses are regulated by plant hormones including salicylic acid (SA) and jasmonic acid (JA). Sometimes, plant defences can be complemented by the presence of symbiotic microorganisms. A remarkable example of this are grasses establishing symbiotic associations with Epichloë fungal endophytes. We studied the level of resistance provided by the grass’ defence hormones, and that provided by Epichloë fungal endophytes, against an introduced herbivore aphid. These fungi protect their hosts against herbivores by producing bioactive alkaloids. We hypothesized that either the presence of fungal endophytes or the induction of the plant salicylic acid (SA) defence pathway would enhance the level of resistance of the grass to the aphid. Methods. Lolium multiflorum plants, with and without the fungal endophyte Epichloë occultans, were subjected to an exogenous application of SA followed by a challenge with the aphid, Sipha maydis. Results. Our results indicate that neither the presence of E. occultans nor the induction of the plant’s SA pathway regulate S. maydis populations. However, endophytesymbiotic plants may have been more tolerant to the aphid feeding because these plants produced more aboveground biomass. We suggest that this insect insensitivity could be explained by a combination between the ineffectiveness of the specific alkaloids produced by E. occultans in controlling S. maydis aphids and the capacity of this herbivore to deal with hormone-dependent defences of L. multiflorum.Fil: Bastias, Daniel.     Jasmonic acid regulation of the anti-herbivory mechanism conferred by fungal endophytes in grasses

    The most studied mechanism of protection against herbivores in grasses associated with Epichloë fungal endophytes has been the fungal production of alkaloids. However, the contribution of the plant immune response on the level of resistance to herbivores in symbiotic grasses has been poorly explored. We studied the relationship between the plant hormone, jasmonic acid (JA) and Epichloë fungal endophytes on herbivore defences in symbiotic grasses. We hypothesized that an exogenous application of methyl jasmonate (MeJA), an activator of the plant JA defence response, would increase the level of resistance of endophyte-symbiotic and non-symbiotic plants to a chewing insect. As Epichloë endophytes produce alkaloids, an enhancement of the JA defence would complement the resistance given by these alkaloids. Lolium multiflorum plants symbiotic and non-symbiotic with the endophyte Epichloë occultans were subjected to an exogenous application of MeJA followed by a challenge with the generalist chewing insect Spodoptera frugiperda. We measured the level of plant resistance to chewing insects, and the defences conferred by host plants and fungal endophytes. Symbiotic plants were more resistant to S. frugiperda than their non-symbiotic counterparts. However, despite the fact that the concentration of JA significantly increased in all plants exposure to MeJA, neither endophyte-symbiotic nor non-symbiotic plants showed an enhanced resistance to insects. Unexpectedly, the exposure of endophyte-symbiotic plants to MeJA led to a reduction in the concentration of loline alkaloids (i.e. N-formyllolines and N-acetylnorlolines), consequently decreasing the level of plant resistance to the herbivore. Synthesis. Our results suggest that, rather than complementing the alkaloid-based defence, the jasmonic acid hormone weakens the anti-herbivore mechanism conferred by Epichloë endophytes. The present study highlights that the interaction between the jasmonic acid hormone and the presence of leaf fungal endophytes can be of importance for the effectiveness of the anti-herbivore defences of symbiotic plants.Fil: Bastias Campos, Daniel.     Magnetic Inflation and Stellar Mass. I. Revised Parameters for the Component Stars of the Kepler Low-mass Eclipsing Binary T-Cyg1-12664

    Several low-mass eclipsing binary stars show larger than expected radii for their measured mass, metallicity, and age. One proposed mechanism for this radius inflation involves inhibited internal convection and starspots caused by strong magnetic fields. One particular eclipsing binary, T-Cyg1-12664, has proven confounding to this scenario. Çakırlı et al. measured a radius for the secondary component that is twice as large as model predictions for stars with the same mass and age, but a primary mass that is consistent with predictions. Iglesias-Marzoa et al. independently measured the radii and masses of the component stars and found that the radius of the secondary is not in fact inflated with respect to models, but that the primary is, which is consistent with the inhibited convection scenario. However, in their mass determinations, Iglesias-Marzoa et al. lacked independent radial velocity measurements for the secondary component due to the star's faintness at optical wavelengths. The secondary component is especially interesting, as its purported mass is near the transition from partially convective to a fully convective interior. In this article, we independently determined the masses and radii of the component stars of T-Cyg1-12664 using archival Kepler data and radial velocity measurements of both component stars obtained with IGRINS on the Discovery Channel Telescope and NIRSPEC and HIRES on the Keck Telescopes. We show that neither of the component stars is inflated with respect to models. Our results are broadly consistent with modern stellar evolutionary models for main-sequence M dwarf stars and do not require inhibited convection by magnetic fields to account for the stellar radii

    Longitudinal white matter changes associated with cognitive training

    Improvements in behavior are known to be accompanied by both structural and functional changes in the brain. However, whether those changes lead to more general improvements, beyond the behavior being trained, remains a contentious issue. We investigated whether training on one of two cognitive tasks would lead to either near transfer (that is, improvements on a quantifiably similar task) or far transfer (that is, improvements on a quantifiably different task), and furthermore, if such changes did occur, what the underlying neural mechanisms might be. Healthy adults (n = 16, 15 females) trained on either a verbal inhibitory control task or a visuospatial working memory task for 4 weeks, over the course of which they received five diffusion tensor imaging scans. Two additional tasks served as measures of near and far transfer. Behaviorally, participants improved on the task that they trained on, but did not improve on cognitively similar tests (near transfer), nor cognitively dissimilar tests (far transfer). Extensive changes to white matter microstructure were observed, with verbal inhibitory control training leading to changes in a left-lateralized network of frontotemporal and occipitofrontal tracts, and visuospatial working memory training leading to changes in right-lateralized frontoparietal tracts. Very little overlap was observed in changes between the two training groups. On the basis of these results, we suggest that near and far transfer were not observed because the changes in white matter tracts associated with training on each task are almost entirely nonoverlapping with, and therefore afford no advantages for, the untrained tasks
